The 25-year-old scored the opener on the stroke of halftime to give the Cottagers a 1-0 lead before the break.
Manchester United captain Bruno Fernandes equalized in the 71st minute to force the game into extra time and eventually the penalty shootouts.
Fulham won 4-3 in the shootouts to progress to the quarterfinals after Victor Lindelof and Joshua Zirkzee missed from the spot.
“It’s a privilege and an honour to be here. To come here, it is a blessing, so I’m happy,” Bassey told BBC.
“I like to say I’m a bit of a poacher. Trying to smell where the ball would go and reacting first. Put my head on it and back of the net.
“They’re a very good team with a lot of talent. We tried to take the sting out of the game and control possession.”
In a post on X, the Nigerian international wrote, “Theatre of dreams.”
It was Bassey’s first goal in the FA Cup.
